Hall of Fame
It's a club of open era players with 25+ wins against their rivals. Only players with GS titles and/or players with top3 career high are included.
So far, only Becker and Djokovic are members.
1. Djokovic-Nadal 26-23
2. Djokovic-Murray 25-11
3. Becker-Edberg 25-10

Whch rivalry will be nr.4?
Who is next to join these 2 guys in this kind of exclusive club?

Is there a chance for Nadal? He needs only 2 wins against Novak or Fed to reach exclusive membership?

He could do it against old man Fed but both are so vulnerable right now they might not even play each other if they were drawn in the same quarter.

I don't think he will beat Djokovic more than once in his career going forward unless they play a bunch of tournaments of clay and Djokovic is out of form/declines a lot. Nadal has beaten Djokovic only once since the 2013 US Open (imagine someone making a prediction of this sort straight after that final!) and bar the 2014 FO final he's 0-11 in matches and 1-22 in sets.

Incidentally Djokovic also has at least 25 wins over Berdych but Tomas' highest ranking was #4 so I can understand why the OP hasn't given him membership to such an elite club. ;)

Yeah, you definitely can't say Djokovic/Berdych is "great rivalry". Still, it's nice trivia. :)

25-2 vs Berdy! Crikey, that surely must be the most lopsided H2H in ATP history!!! It's no wonder the Birdman loses a match vs Djoko the moment he steps on court, you could see from his body language.

I'd argue that a lopsided rivalry is not great. A 75% or greater win rate against another player should not qualify as a great rivalry, but incredibly close, often matches should.

Hence, I believe the criteria here should be:
  • At least 20 matches played.
  • The more dominant player should have no more than 75% of the wins
And to be an all-time great rivalry:
  • Both of the above
  • Both players should be in the top 15 throughout 80% of the rivalry
So Federer-Djokovic is an all-time great rivalry, particularly as the H2H is so close.
Berdych-Djokovic is not, because Djokovic wins 92.6% of the matches.
